1.闡明孤獨(dú)癥關(guān)鍵致病因子的功能及致病機(jī)制 | 通過自主構(gòu)建孤獨(dú)癥關(guān)鍵致病因子的基因編輯動物模型,揭示了FOXG1、Neurexin/Neuroligin、ROCK/PAK/LIMK等致病因子或信號通路在大腦模式形成、神經(jīng)元命運(yùn)決定和發(fā)生、神經(jīng)元遷移和軸突投射、突觸形成和通常可塑性等過程中的作用,揭示了上述致病因子和信號通路異常導(dǎo)致孤獨(dú)癥的機(jī)制,對闡明兒童孤獨(dú)癥的發(fā)病機(jī)制及相應(yīng)診治具有重要意義。 | ||||||
2. 多基因協(xié)同促進(jìn)內(nèi)耳干細(xì)胞再生重建聽覺功能 | 研究了Wnt、FoxG1、Spectrin等基因在內(nèi)耳干細(xì)胞增殖分化、毛細(xì)胞成熟和存活中的作用機(jī)制;優(yōu)化了高效轉(zhuǎn)染內(nèi)耳干細(xì)胞的新型腺相關(guān)病毒, 建立了小鼠內(nèi)耳干細(xì)胞的基因治療系統(tǒng)。研究了多種生物材料和電、磁等物理刺激對聽覺神經(jīng)元再生、功能成熟和存活的調(diào)控機(jī)制。開創(chuàng)性地研究了自噬和免疫系統(tǒng)促進(jìn)毛細(xì)胞和神經(jīng)元存活的作用和機(jī)制。 | ||||||
3.揭示本能行為和學(xué)習(xí)記憶的分子與神經(jīng)環(huán)路機(jī)制 | 闡明了生物節(jié)律通過特異性節(jié)律輸出分子Fbxl4調(diào)控睡眠的分子細(xì)胞機(jī)制,揭示生物節(jié)律調(diào)控睡眠行為的作用機(jī)制。闡明機(jī)體在睡眠和求偶兩種本能行為間抉擇的機(jī)制,揭示了睡眠與求偶行為在神經(jīng)環(huán)路上的相互作用。發(fā)現(xiàn)PKCλ與PKMζ分別在學(xué)習(xí)記憶的早期表達(dá)、鞏固及后期維持階段的作用,揭示一種存在于胞體的調(diào)節(jié)突觸傳入的細(xì)胞新機(jī)制。 | ||||||
4.揭示干細(xì)胞命運(yùn)決定及重編程的轉(zhuǎn)錄調(diào)控機(jī)制 | 發(fā)現(xiàn)鋅指蛋白ZFP281募集AFF3到增強(qiáng)子上精確調(diào)控印記基因的差異性表達(dá),以及協(xié)同甲基轉(zhuǎn)移酶MLL2調(diào)控轉(zhuǎn)座子LINE-1區(qū)的組蛋白修飾以及染色質(zhì)局部結(jié)構(gòu),調(diào)控干細(xì)胞的命運(yùn)決定及重編程。揭示SEC介導(dǎo)多價相分離調(diào)控RNA聚合酶II轉(zhuǎn)錄暫停釋放,從全新角度闡明基因快速協(xié)調(diào)轉(zhuǎn)錄激活程序的機(jī)制,為探索SEC相關(guān)疾病的機(jī)制及篩選靶向藥物提供了新思路。 | ||||||
5. 原發(fā)性膽汁性膽管炎發(fā)病機(jī)制的研究 | 通過對漢族人群原發(fā)性膽汁性膽管炎(PBC)易感基因的研究,在國際上首次發(fā)現(xiàn)IL-21信號通路和T細(xì)胞輔助激活的異常在PBC發(fā)病機(jī)制中的重要作用,并通過CD58易感基因的發(fā)現(xiàn)把膽汁上皮細(xì)胞相關(guān)基因功能和PBC發(fā)病機(jī)制結(jié)合起來,為進(jìn)一步探討PBC發(fā)病機(jī)制提供了線索。 | ||||||
